The story of Manyam Puli begins with the childhood days of Kumar (Mohanal), his mother dies after giving birth to his brother Manikuttan (Vinu Mohan), and is left with his brother, his father (Santhosh Keezhattoor) and His uncle Balaraman (Lal). A few days after his mother's death, he also lost his father in a tiger attack, as they escaped from the police illegally cutting logs in the woods. Kumar in revenge traps the predator with the help of Balaraman and kills him. He soon became the savior for the natives of Puliyur, who feared frequent tiger attacks in the village.
After many years, Murugan is now a truck driver by profession, he has a housewife, Myna (Kamalini Mukherjee) and a daughter, Chakki (Baby Durga Premjith). He is an active rogue tiger hunter in Puliyoor, whenever he endangers the village. Apart from the combat sequences with the tigers / leopards, the film also tells the moments of the family of Kumar (Mohanlal) in the context of a history of revenge.
